This program discusses and illustrates with the aid of graphs, charts, and drawings the various factors affecting electrophoresis. Emphasis is placed on the fact that these factors interact and must be looked at critically when designing a procedure for electrophoresis. Initially, the principle in physics upon which electrophoresis is based is identified. Factors to consider in the selection of buffers, and the mixture of different protein molecules are discussed next. The advantages and disadvantages of filter paper versus those of cellulose acetate are then noted. In addition, the parameters influencing electrophoresis, namely, the ionic strength of the buffer, pH of the buffer, voltage level, current level, running time, electroendosmosis, evaporation and wick flow are discussed. Two methods for quantitation of the electrophoretic separations are also identified. One uses a spectrophotometer and the other one a densitometer
