A second mutant of Escherchia coli B (strain RD-2) with impaired potassium transport has been isolated with nitroguanidine as the mutagenic agent. Except for its potassium requirement, RD-2 manifests the same nutritional requirements as the parent strain and contrasts with RD-1, which was shown to be deficient both in potassium transport and in methionine biosynthesis. RD-2 will not grow in a low potassium growth media which supports growth of the parent strain. Potassium accumulation studies demonstrated that the mutant low potassium growth failure is attributable to a potassium accumulation impairment

Studies with potassium transport mutant, strain RD-2, reveal that the mutant does not take up inorganic phosphorus normally. The abnormality in phosphorus uptake is reflected in a reduced rate of 32Pi incorporation into phospholipid. The 32Pi uptake reaction of the mutant is super-stimulated by potassium and inhibited by sodium, characteristics which resemble the behavior of the potassium-stimulated enzymes that are most commonly found catalyzing reactions in intermediate metabolism that involve the transfer of a phosphate group

Pulsed nuclear magnetic resonance (NMR) measurements of K39 in solutions and bacteria were made, by the use of a super-conducting magnet with a field of 50,300 gauss, which provides the necessary increase in sensitivity. Transverse relaxation time (T2) of K39 in a 1 M KC1 solution showed a mean value of approximately 0.0008 seconds. T2 of K39 in aqueous solutions increased with increasing concentrations of potassium. Fifty-nine per cent of K39 in packed wet ion exchange resin was invisible to NMR, presumably due to a marked shortening of T2 complexed by the resin. T2 of K39 in the bacterium H. halobium was shortened by at least 50 per cent, which suggests that cell K(+) is not in free solution in liquid water, but may be complexed to fixed charge sites or solvated in semicrystalline water of the cell.
