The effect of several hydrazine and nonhydrazine compounds on the concentrations of norepinephrine and histamine in the rat heart was studied. The compounds used were either monoamine oxidase (MAO) or diamine oxidase (DAO) inhibitors. Hydrazine (0.31 mmole/kg.), monomethylhydrazine (0.31 mmole/kg.), pargyline (0.16 mmole/kg.), and tranylcypromine (0.16 mmole/kg.) produced significant elevations of norepinephrine in the heart. The increase produced by monomethylhydrazine (MMH), pargyline, and tranylcypromine may be due to the fact that they are potent MAO inhibitors, but this does not explain the effect of hydrazine which is devoid of any MAO inhibitory action. Histamine levels in the heart were elevated only by MMH (0.31 mmole/kg.) and pargyline (0.16 mmole/kg.). MMH inhibited DAO, but pargyline does not have any effect on this enzyme. The mechanism whereby hydrazine increased norepinephrine in heart and pargyline elevated histamine does not appear to be dependent on their ability to inhibit the enzymes connected with the metabolism of these two amines. These experiments indicate that the extent of MAO or DAO inhibition by a specific compound is not related to its ability to affect tissue amine levels. The data also show that exposure to MMH or hydrazine could affect the cardiovascular system by changing the levels of amines in the heart

Effects of Monoamine Oxidase Inhibitors and Reserpine on Brain Amines in Altitude-Exposed Rats by James H Merritt( Book )

Groups of rats were injected with either pargyline, Parnate, Catron, or reserpine (75 mg./kg., 5 mg./kg., 5 mg./kg., and 5 mg./kg. respectively) and taken to a simulated altitude of 18,500 ft. The expected rise in the brain monoamine (norepinephrine and serotonin) levels after administration of monoamine oxidase inhibitors and the decrease of norepinephrine by reserpine were attenuated by altitude exposure. In another experiment, 100 percent oxygen was substituted for ambient air. In this case, the attenuation of monoamine elevations by the inhibitors was reversed. However, the added oxygen did not reverse the attenuation of the norepinephrine decrease by reserpine. (Author)
Effects of decaborane on liver glycogen content of rats at ground level and at altitude by Ralph E Grandberry( Book )

The effect of decaborane (15 mg./kg.) on the hepatic glycogen of male rats maintained at altitude (18,000 ft.) under normoxic conditions was compared with the effect on similarly injected, ground level controls. Results showed that decaborane decreased the glycogen level of fed rats at ground level, but did not affect glycogen levels in the fed, altitude group. The hepatic glycogen level of fasted rats at altitude was the equivalent of that of fasted rats at ground level. These results indicate that the mechanism whereby decaborane produces a depletion of liver glycogen is altered under conditions of reduced pressure

Effects of arginine, l-glutamate, and pyridoxine on the toxicity of Aerozine by Miguel Angel Medina( Book )

The toxicity of the rocket propellant Aerozine, a mixture of hydrazine and 1,1'-dimethylhydrazine (UDMH), was investigated in rats. It was found that Aerozein has an LD 50 of approximately 120 mg/kg and produced severe convulsions within 30 minutes after intraperitoneal injection. Administration of a mixture arginine-glutamate (4 mM/kg) and pyridoxine (0.5mM/kg) produced a significant reduction of deaths in rats given 130 mg/kg of Aerozine. This antidotal combination was not as effective when glutamate was omitted, while injections of arginine-glutamate, glutamate-pyridoxine, arginine, l-glutamate, or pyridoxine alone did not afford any protection. These experiments indicate that the combination of arginine-glutamate and pyridoxine may be of therapeutic value in the event of a human exposure to Aerozine
Inhibition of D-aspartate uptake in rat cerebral cortex slices by hydrazines by Miguel Angel Medina( Book )

The uptake of D-aspartate by rat cerebral cortex slices was found to consist of two processes: an energy-requiring system, and possibly nonspecific binding. The accumulation of D-aspartate was inhibited by specific concentrations of monomethylhydrazine (MMH), and 1,1-dimethylhydrazine (UDMH). At higher concentrations, hydrazine inhibited uptake by 50 %, while UMDH produced a 25% inhibition. Uptake of D-aspartate in cerebral cortex slices from rats given UDMH and sacrificed before any convulsions was no different from control values. The accumulation of D-aspartate in cerebral cortex slices of rats given the same dose of UDMH but allowed to convulse was greater than that of the controls. The lack of inhibition of uptake in cerebral cortex slices from UMDH-injected animals militates against the suggestion that UDMH might produce convulsions by interfering with active transport across neuronal membranes

The objective of this research is to develop a groundwater remediation advisory system for use in investigating possible remediation activities for the cleanup of contamination from hazardous substances, pollutants and contaminants at Air Force sites. The advisory system provides a decision framework for selecting an appropriate model from the numerous available transport models conditioned on available site data. In addition, the use of optimization methods is explored for determining optimal remediation strategies at a specific site. The optimization routine evaluates tradeoffs between the long-term cost of remediation and the probability the remediation strategy will fail. The final product of this project is a computer-based Air Force Installation Restoration Workstation Advisory System for contaminant modeling and decision-making. This software can be used as an aid to technical project managers within the U.S. Air Force Installation Restoration Program in developing and evaluating possible remediation alternatives and managing ongoing remediation activities.
