Cytokines and cytokine receptors remain an area of great interest for the development of targeted therapies for cutaneous inflammatory diseases. Anti-TNF therapeutics have proven to be effective in the treatment of psoriasis, and clinical investigations have now begun for other cytokine-directed therapies, such as those targeting IFN-g, IL-12p40, and IL-18. In addition to therapeutics that target cytokines directly, strategies that target cytokine signaling pathways are in development. This book summarizes the findings of the 56th International Workshop of the Ernst Schering Research Foundation that focused on "Cytokines as Potential Therapeutic Targets for Inflammatory Skin Diseases."

Preliminary experiments outlined in the original application suggested that supernatants from mononuclear cell cultures stimulated with staphylococcal antigens or IL-2 could release SRIF from dispersed cortical neurons. Such activated supernatants are known to contain many cytokines, including interleukins-1 (IL-1). We elected to begin the study of this release activity by examining the effects of IL-1 and IL-2 in more detail, using pure products of recombinant DNA origin. Human recombinant interleukin-1-beta (rIL-1B) was tested in both cortical and diencephalic cells. Modest SRIF release activity was observed only at a concentration of 10- M. To investigate the effect of chronic IL-1 exposure, cultures were incubated for 48 hours with various concentrations of rIL-1B and the cells and media assayed for SRIF. Keywords: Somatostatin

Cytokines are polypeptides synthesized by many cells which act on a variety of tissues by changing gene expression and cellular metabolism. The pro-inflammatory cytokines are a particular group of cytokines with molecular weights between 8,000 - 25,000 Daltons which appear to be synthesized primarily in association with disease states or during host perterbation. Some 'normal' physiologic events such as strenuous exercise or normal ovulation result in the synthesis of the pro-inflammatory cytokines. These polypeptides are very potent molecules which, at picomolar or even femtomolar concentrations, trigger a variety of responses in cells in vitro but similar concentrations may be effective in vivo when one considers the half-life, clearance and distribution of parenterally administered cytokines

Las infecciones por bacterias gram negativas son una de las primeras causas de muerte en el recien nacido. La depuracion de bacterias es deficiente en el neonato, situacion que aumenta la susceptibilidad a las infecciones. En este estudio se logro mejorar el patron de depuracion de Klebsiella pneumoniae en ratas wistar recien nacidas inoculadas por via IP con 800mg/k de fosfatidilcolina de soya (FC), en contraste con el grupo testigo inyectado con PBS(p<0.05). La sobrevida de los animales aumento (p<0.05), y los cambios leucocitarios se caracterizaron por una mayor leucocitosis y neutrofilia durante el pico de bacteremia en los animales tratados con FC. Los niveles circulantes de interleucina-6 fueron mayores en el grupo de FC, observandose ademas hematopoyesis extramedular de las series granulociticas (p<0.05) y de megacariocitos (p<0.01) en el bazo de las ratas del grupo de FC. No se observaron cambios significativos en los depositos de granulocitos en la medula osea de ambos grupos. La mejoria de la sobrevida, los cambios leucocitarios y los focos de hematopoyesis extramedular, parecen asociarse al aumento en la produccion de IL-6. Los resultados sugieren que la IL-6 participa en el mecanismo de proteccion; y que la FC induce en este modelo a septicemia experimental. La FC parece comportarse como un inmunomodulador inespecifico de la respuesta aguda a la infeccion bacteriana(AU)

(CIT): 2010 Kinyoun Lecture Dr. Charles Dinarello, who purified and cloned interleukin-1beta (IL 1beta), once known as the fever molecule, will deliver the 2010 Kinyoun Lecture on Thursday, Nov. 18, at 3 p.m. in Lipsett Amphitheater, Bldg. 10. Dinarello is professor of medicine and immunology at the University of Colorado School of Medicine and a former investigator in NIAID"s Laboratory of Clinical Investigation. Dinarello"s talk, titled "Treating Auto-inflammatory Diseases with Interleukin-1 beta Blockade," will include an overview of autoinflammatory diseases, such as type 2 diabetes mellitus and gout, and describe differences between these and autoimmune diseases. He also will discuss treatment of inflammatory diseases using IL-1beta blocking agents. Considered one of the founding fathers of cytokine biology, Dinarello"s work with IL-1 helped to establish the significance of cytokines in inflammatory diseases. His recognition of the importance of IL-1 in inflammatory diseases led to the development of therapies that inhibit IL-1 and thereby reduce disease severity. These therapies include those based on the IL-1 receptor antagonist, monoclonal antibodies and soluble receptors
