Cette thèse a été consacrée à la synthèse de nouveaux dérivés d'acides pulviniques et à l'évaluation de leurs propriétés antioxydantes, pour développer de nouveaux composés à incorporer dans des crèmes à usage dermocosmétique. Les nouveaux composés sont des dérivés dits mono-aromatiques d'acides pulviniques, ne possédant qu'un seul noyau aromatique, soit sur la partie "sud" (produits de type I), soit sur la partie "nord" (produits de type II). Des voies de synthèse ont été développées et appliquées à la préparation de 45 nouveaux dérivés. L'activité antioxydante de ces composés a été évaluée et comparée aux acides pulviniques classiques. Les tests de protection de la thymidine et d'un plasmide bactérien, ont permis de montrer que les composés de type I étaient aussi actifs que les acides pulviniques classiques, contrairement aux produits de type II, peu actifs. Des tests complémentaires ont montré l'importance de la fonction portée par la double liaison exo-cyclique dans l'activité

The objective of this thesis was to synthesize and to evaluate the biological activity of hydrophilic derivatives and structural analogues of pulvinic acid, derived from norbadione A, a naturally occurring antioxidant found in mushrooms. This study aimed to develop new protective agents against ionizing radiations. First a scalable synthesis of a bis-lactone as a common intermediate was developed. It is based on the condensation and dehydratation of the anion of a protected tetronic acid on methyl pyruvate and subsequent selective demethylation in presence of magnesium bromide. This intermediate was then employed to prepare several hydrophilic derivatives by regioselective ring-opening of one lactone by functionalized amines. The second study was based on the creation of an in silico activity-predicting model allowed to highlights derivatives of 3-aryl-4-hydroxycoumarines as potential antioxidants. A synthesis pathway was used to prepare several derivatives to perform a structure-activity relationship study. All synthesized compounds were then tested in an in vitro assay, to investigate their radioprotective properties. Radiosensitive cells were exposed to the different compounds and irradiated, after several days their viability was assessed. Finally a third study was conducted to investigate the chelating activity of pulvinic acids towards 137Cs in order to develop new detoxifying agents

Nanoparticles are extensively studied in the medical area either for the delivery of active principles or for the diagnostic. They are essential in the development of new material thanks to their unique properties. So, we have decided to work on three kinds of nanostructures made from new polymerizable self-arrangements of diacetylenic amphiphiles: spherical micelles, polymerized lipidic constructions adsorbed on carbon nanotubes surface and nanotubules of lipids. First, we have tagged spherical micelles by encapsulation of a fluorescent dye. Then, the potential of these micelles as delivery agent has been established on HeLa cells. In addition to that, technicium has been chelated on the micelles surface to study their biodistribution and their evolution in the blood flow. In a second time, several amphiphiles have been arranged around carbon nanotubes in the aim to forecast the stability of carbon nanotubes dispersions at different pH. To end, the discovery of nanotubules coming from self-arrangements of a specific amphiphile have been studied to elucidate their structure. Indeed, we showed that these nanotubules were constituted of a belayed of amphiphile which self-arrange in ribbons, then in helix and tubes

Today, organocatalysis is one of the most important branches of the asymmetric synthesis along with the enzymatic and the metallic catalysis. We were particularly interested in this field while developing a retrosynthesis of a natural product, the Nigellamine A. In fact, the key step relies on a direct intramolecular aldol reaction of an acyclic ketoaldehyde. We therefore developed and used guanidines as potential organocatalysts for this transformation. A consequent study allowed the formation of ketol compounds from ketoaldehydes with excellent chemoselectivity and good yields. We then applied this methodology to the total synthesis of the Nigellamine A, an active compound with hypolipidemic properties. The cyclization was not as chemoselective as expected and the purification of the desired ketol by silica gel chromatography was not efficient and needs to be optimized. In parallel, we also used chiral guanidines as catalysts for the enantioselective acetylation of amines. Finally, the last part highlights a new methodology developed for the synthesis of [alpha]-alkylated butenolids, important precursors for many natural products, and its application to the total synthesis of (+)-Ancepsenolide

The first part of this paper deals with a high-throughput procedure for crude mixtures capable of rapidly analysing a mixture whatever its composition. This method uses mass spectrometry to detect substrates functionalized with a tag, chemically inert and with specific characteristics. After having defined the tag structure (chemical stability, reaction inertia, ionisation coefficient...), this process was used to evaluate the influence of operating conditions on several coupling reactions, to improve a palladium-catalysed decarboxylative coupling, to highlight an unexpected reaction, and, lastly, to see how tagged natural products evolve in a crude mixture. The second part of this paper sets out the three strategies used in the industrial production of (2S,3R,4S)-4-hydroxyisoleucine, a natural product with anitdiabetic properties. The first pathway is based on an enantioselective reduction of a racemic precursor using ruthenium catalysts in the presence of chiral ligands such as 1,2-diamine. The second pathway is based on the diastereoselective reduction of an achiral isoxazole intermediate to obtain a racemic mixture of a diastereoisomerically enriched 4-hydroxyisoleucine. The last pathway is based on a proline-catalysed asymmetric Mannich condensation which produces (2S,3R,4S)-4-hydroxyisoleucine without any chromatographic purification and with a global yield of 22%

Molecular imprinting is a technique that enables the preparation of synthetic receptors in a highly cross-linked polymer matrix. This technique has known a remarkable development in recent decades, and has currently a large number of potential applications, such as stationary phase for chromatographic columns, as a support in solid phase extraction (SPE), as nanoreactors and as catalysts. During my thesis work, we developed a new method for the preparation of surface imprinted nanospheres, combining for the first time the semi-covalent approach with the miniemulsion polymerization. As a first step, we wanted to assess the parameters that influence the selectivity of molecular imprinted polymers (MIPs) prepared with this new imprinting technique. We prepared different imprinted polymers with an increasing number of carboxylic acid functions in the binding sites, using phosphorous templates. This increase leads to the generation of an artificial receptor with greater selectivity for the recognition of original analytes. Then, we applied the new imprinting technique to the preparation of artificial receptors for glucose derivative. The imprinted polymer showed an excellent imprinted factor and a remarkable selectivity for glucose, compared to other epimers. The selectivity factor found between glucopyranoside and galactopyranoside is the highest reported so far

Over the last decades, organometallic chemistry and catalysis contributed to the progress of modern organic chemistry, affording new synthetic methods used not only for the synthesis in laboratory, but also in industry. The presented work deals with three different organometallic reactions leading to key molecular motifs: mandelic esters, propargylic alcohols and phosphinoyl amides. In the first part we have thus presented the cross coupling reaction between aromatic boronic acids and ethyl glyoxylate as a catalytic approach to alpha-hydroxy esters. We also discussed the results obtained with enantio- and diastereoselective strategies. The second part treats the addition of lithium alkynylborates, as a truly original and chemoselective approach toward the synthesis of highly functionalized propargyl alcohols. In the last part of the manuscript we have introduced the synthesis of new P-chirogenic or chiral for the inducer derived from TADDOL directly linked to the P atom, N-phosphinoyl imines. The evaluation of the diastereoselectivity derived from the chiral inducer close to the C, N double bond have been done by the 1,2 addition of organometallic reagents
Marquage métabolique de glycanes : diagnostic et approches thérapeutiques by Laura Fourmois( )

This PhD work focuses on glycans metabolic labeling. This method uses a modified monosaccharides bearing a chemical reporter. The unnatural monosaccharide is metabolically incorporated into glycans. Two targets were selected, Legionella pneumophila bacteria and prostate cancer cells (PC3).Various saccharidic analogs were synthesized carrying several chemical reporters, like azide, terminal alkyne, terminal alkene, cyclopropene and ketone functions. A common synthetic strategy was developed using N-hydrosuccinimide ester derivatives and amino monosaccharides, such as D-mannosamine, D-galactosamine and L-fucose for eukaryotic cells and analogs of a legionaminic acid precursor for Legionella pneumophila bacteria.Differents metabolic labeling of glycans were carried out on Legionella pneumophila. Various sugar derivatives were incorporated by bacteria, then the reporter group was reacted selectively and covalently with a complementary function (azide-cyclooctyne, alkene-tetrazine, ketone-hydrazide/alkoxyamine). Bacteria were visualized with an imaging probes by light microscopy (directly: partner bearing a fluorophore, indirectly: recognition of a biotin group by a fluorescent streptavidin). The results highlighted outer membrane labeling.For eukaryotic cells (PC3), metabolic oligosaccharides engineering has been accomplished to check chemical reporter analogs incorporation via detection with fluorescent probes. A series of tools carrying functions complementary to the chemical reporters was synthesized, such as cyclooctyne derivatives (TMDIBO) and tetrazine derivatives. These derivatives were combined with biotin groups for detecting tools or with Antibody Recruiting Molecules (ARMs), such as 2,4-dinitrophenyl and L-rhamnose, for potential therapeutic approaches. The concept focuses on the combination of metabolic glycan labeling and the activation of human serum complement by ARMs to kill selectively labeled cells. First, ARMs labeled cells was checked by recognition of fluorescent anti-ARMs antibodies. Then, metabolic glycan labeling and human serum complement activation has been evaluated but the complement activation protocols still need to be optimized.Metabolic labeling of glycans is an effective method to detect, by fluorescence, Legionella pneumophila bacteria using analogs of a legionaminic acid precursor and eukaryotic cells (PC3) with monosaccharide derivatives. The labeling was observed on the membrane of the two targets. This method can potentially combine imaging or diagnostic aspects with various therapeutic approaches
