(R)-(+)-[beta]-Benzyl-[gamma]-butyrolactones are key-intermediates in the total syntheses of natural lignans and analogues. The only optically active lactone of this type, which was known before us, is (R)-(+)-[beta] piperonyl-[gamma] -butyrolactone, two syntheses of which have been described in the literature. Both these syntheses being long and consequently of limited preparative value, we sought to develop a simpler and more efficient route leading to optically active [beta]-benzyl-[gamma]-butyrolactones. Thus, the methyl (R,S)-[alpha]-benzylhemisuccinate resulting from a STOBBE condensation with an appropriate aromatic aldehyde, followed by catalytic hydrogenation of the intermediate [alpha]-benzylidene hemisuccinic ester, was resolved by means of a chiral base. Reduction of each enantiomer, using calcium borohydride, then led to the corresponding optically active [beta]-benzyl-[gamma]-butyrolactone. In this way, we obtained the following three lactones in both (R)-(+) and (S)-(-) enantiomeric forms, [beta]-piperonyl-, [beta]-veratryl-, and [beta]-vanillyl-[gamma]-butyrolactones. We used these chiral precursors to synthesize various lignoids including fifteen natural lignans

L'exposé décrit dans sa première partie l'étude chimique d'un corail mou, Xénia membranacea, récolté en Nouvelle Calédonie. Vingt-sept nouveaux diterpènes du type xénicine ont été isolés et caractérisés ainsi que trois stérols connus. La configuration absolue du constituant majoritaire, la havannahine et de quatre de ses dérivés, a été déterminée par corrélation chimique et analyse par diffraction des R.X. Une étude approfondie par RMN1H et 13C à deux dimensions a permis de discuter et de déterminer la configuration relative des trois désoxy-11,19 havannahines. La deuxième partie soutenue à huis clos décrit des approches de synthèse stéréosélective de dérivés d'amino-2'imidazole isolés d'organismes marins

We have studied the eniymatic mechanisms involved in the bioconversion of glyceryl trinitrate (GTN) by a filamentous fungus : Phanerochaete chrysosporium and in the GTN metabolization by hepatic cells extract . The following resuits have been pointed out - The fungus degrade GTN in 2-GMN with a rate of 90% and 80% of regioselectivity. EPR spectra made during GTN incubation show the formation of intracellular complexes identified like hemoprotein-NO and protein Fe-S-NO. - The GTN degradation by P. chrysosporium involved enzymatic systems_ constituted by soluble and membrane activities. The cytosolic activity is constituted by a glutathion-transferase and by an hemoprotein like cytochrome P-450. Hemoproteins seems to be involve in membrane activities and in particular a cytochrome P-450 in microsomal fraction. - Hepatic rat microsomes, induced by phenobarbital, interact with GTN to form a type I spectrum. In presence of NADPH and in anaerobic condition, a spectrum appeared, caracteristic of an interaction cytochrome P-450-NO. These microsomes degrade GTN in di and mononitrated derivatives. Amongst the cytochrome P-450 inducer tested, dexamethasone increases stongly the concentration of cytochrome P-450 involved in the reductive degradation of GTN. This result and the effect of specific inhibitors allow us to involve the IIIA form of cytochrome P-450
