70Z/3 is a murine pre-B cell leukemia line that has been used extensively in the study of signalling pathways in B cells. As a tumour, 70Z/3 cells were initially found to cause widespread disease upon injections in animals. We have isolated 70Z/3 variants divergent in their capacity to lead to morbidity after injections. One variant, 70Z/3-NL, elicits an immune response capable of eliminating the tumour. Another variant, 70Z/3-L, is not rejected and causes morbidity. I found that an immune response generated against 70Z/3-NL cells protects against a challenge with 70Z/3-L cells. I further observed, through the isolation of subclones from 70Z/3-NL, that immune recognition of only a portion of the cells is sufficient to protect against the tumour. Finally, I used 70Z/3-L to test an anti-tumour therapy based on the use of low doses of IL-12. I established a protective therapy against an intraperitoneal challenge with 70Z/3-L cells with doses of IL-12 ranging from 500 to 1000 ng/kg. IFN-gamma has been associated in many IL-12-based therapies with both therapeutic effects and toxicity. This protocol of daily injections of IL-12 led to protection of challenged animals but did not induce IFN-gamma when mice received only injections of IL-12. Serum levels of IFN-gamma increased only when both tumour cells and IL-12 were administered to the animals. I determined that CD4+ and CD8+ T cells were required for the IL-12 therapy while NK cells were not necessary. I conclude that we can use a dose of IL-12 that does not directly induce IFN-gamma. In this context, although IFN-gamma is ultimately needed for the rejection, it is not an initiating event of low dose IL-12 therapy. The ability of 70Z/3-NL cells to initiate immune rejection is of clinical relevance, especially if we can find the gene, or genes, responsible for the initiation of the immune response. I have used both serial analysis of gene expression (SAGE) and microarrays to compare gene expression between 70Z/3-NL and 70Z/3-L.I have confirmed differential expression of two genes, TSA-1 and SPARC

Cette étude porte sur la métamorphose dans la littérature narrative médiévale de langue d'oïl (XIIe-XIIIe siècles). Nous cherchons à montrer comment le thème du passage d'un règne à un autre de la nature, issu du fonds païen mythologique, a été remployé par les auteurs du Moyen Âge. Dans une conception encore païenne du thème, la métamorphose passe par une première adaptation avec la moralisation courtoise des "Métamorphoses" d'Ovide ("Ovidina"); elle peut aussi être considérée comme un instrument de connaissance ou d'initiation à l'amour (la femme-guivre du "Bel Inconnu" et les histoires de loup-garou); dans tous les cas, se pose le problème de l'instance qui la permet (le "mirabilis", le "miraculosus" et l'"humanus"). Dans une perspective chrétienne, elle comprend les opérations diaboliques et celles de Dieu (mises en scènes des dogmes et théophanies, miracles de métamorphose). Le pouvoir de modifier l'ordre de la Création est alors réservé exclusivement à la divinité : le diable, lui, ne peut qu'en produire l'illusion, ses actions se réduisant à des travestissements éphémères. Parallèlement à cette évolution conceptuelle, le thème de la métamorphose connaît une évolution littéraire qui le conduit à une progressive réduction et à la dissolution du phénomène en procédé rhétorique. Au début du XIVe siècle, l'"Ovide moralisé", qui excède le cadre chronologique de notre étude, représente le point final de cette involution

In verse chronicle or novels in medieval French literature, we tend to recall only the exploits of exceptional heroes, mortals whose physical feats one admires or enchanters with enviable qualities. Yet, jealousy and passion, while corrupting bodies through the rigours of combat, give birth to that which becomes an intangible creature in its own right, namely otherness, which, unseen, intrudes between characters in order to harm them and so the better to destroy them. This is an underhand war, conducted according to three perfect strategies. The first entails poisoning relations both too similar and yet too different. Or, it can involve changing a single character - a knight profoundly shaken who no longer knows who he is - himself or another. Finally, otherness, the true dynamic of these works, chooses to attack the victim on both fronts, thus creating turmoil embracing change in oneself and in the other protagonist, there being no chance of knowing which one will prevail
