The human tissue Kallikreins fulfill important regulatory functions in desquamation of epidermal skin layers. Kallikrein 11 (hK11) is strongly expressed in the skin, however the physiological and pathological effect of this protease are rather unclear. In this work hK11 was cloned, brought to secretion in the yeast Pichia methanolica as well as in CHO-cells and applied in activity studies to the determination of kinetic parameters. Moreover, for the first time the inhibitory activities of domains of the serinprotease inhibitor LEKTI on recombinant hK11 has been demonstrated. Domains 6 and 9 exhibited a significant inhibitory effect on hK11 activity. However, other tested domains showed hardly any effect. For LEKTI domain (LD) 6 the Ki-value was determined and the results indicated that LD6 is a mixed noncompetitive inhibitor of hK11

ERp29 ist ein ER-Protein aus der Familie der PDI-Proteine. Literaturhinweise auf seine Funktion in der Zelle basieren auf in vitro-Untersuchungen in der Zellkultur und erwiesen sich zum Teil als widersprüchlich. Als Alternative bieten sich in vivo- Untersuchung an einem ERp29-Knockout (KO)-Maus-Modell an, in dem dieses Protein vollständig eliminiert wurde. Die ERp29-KO-Maus ist das einzige lebensfähige Maus-Modell der PDI-Familie. Die Abwesenheit von ERp29 beeinflusst bei embryonalen Fibroblasten (MEF) weder den Immortalisierungsverlauf noch ihre Immortalisierungsrate. Im Gegensatz zu immortalen Wildtyp (WT)-Zellen reagierten immortalisierte KO-Zellen in Klonogenitäts- und Migrationsstudien sensitiver auf die Behandlung mit einem Karzinogen, zeigten aber keine Anomalien bei der Antwort auf oxidativen Stress. Bei der Analyse der Stressantwort in adulten Zellen (dermale Fibroblasten, Thyreozyten) konnte eine verminderte Aktivität der ATF6-CHOP-Achse der "unfolded protein response"-Kaskade in KO-Zellen festgestellt werden, die zu einer höheren Apoptose-Resistenz im Vergleich zum WT führte. Die Analyse der Proteinexpression in Leber und die primäre Phenotypisierung (in Zusammenarbeit mit der German Mouse Clinic in München) zeigten wesentliche Veränderungen im Fettstoffwechsel der KO-Mäuse, die mit zunehmendem Alter in Fettleibigkeit resultierten. Die ERp29-defizienten Mäuse zeigten im Vergleich zum WT erhöhte Futteraufnahme, verminderte Aktivität und verminderte Reproduktivität. Darüber hinaus konnte bei KO-Mäusen eine verminderte Leptin-Expression im Hypothalamus, eine erhöhte Freisetzung des Stress-Hormons Kortikosteron, eine verminderte Knochendichte sowie Herzrhythmusstörungen festgestellt werden
