Over 800 photographers and publishers created the work represented in the Stereograph Collection. Major publishers include E. & H.T. Anthony, Keystone, Underwood and Underwood and Kilburn Brothers. Photographers include William Henry Jackson, John P. Soule, Carleton Watkins, D. Barnum, Bierstadt Brothers, George Barker, Mathew Brady, Pach Brothers, Napoleon Sarony, J. Gurney and Son, and amateurs F.F. Thompson and Alfred T. Loonam.

Science and technology are represented in the collection, including views of engineering feats such as railroads, aqueducts, canals and bridges. Some early examples of the use of photography to document are included in the views made on the various explorations in the western United States: Fisk's expedition to the Dakotas in 1866, the Geological and Geographical Survey of the Territories (U.S.), led by F.V. Hayden, and the U.S. Topographical and Geological Survey of the Colorado River led by J.W. Powell, as well as images of the full moon.
Many events and activities are also recorded in the collection, including parades, anniversaries of historic events, and holidays such as the Fourth of July. Activities for enlightenment and amusement include rowing, croquet, archery, horseback riding, camp meetings, chatauquas, reading, band concerts, circuses, and picnicking are shown, as well as spectacular events including balloon ascensions, Blondin crossing Niagara Falls on a tightrope, the Chicago and Boston fires, the San Francisco earthquake, train wrecks, dam failures, tornado damage and other disasters. War also offers a perennial subject for photographers, and some of the earliest images in the collection show Crimean War sites. The United States Civil War is heavily documented, and other military actions are recorded, including the Modoc Indian War, the Spanish American War, South African (Boer) War, the Russo-Japanese War, and World War I.
The Stereograph File includes portraits of prominent people of the 19th century, including academics, religious figures, actors and actresses, aristocrats, soldiers and government figures and others. The official visits of the Japanese embassy, the Prince of Wales and the Grand Duke of Russia in the 1860s are all documented. Other foreign-born individuals are identified, including Chinese laborers in Massachusetts and California, and Irish-, German-, and Scandinavian Americans. African Americans are depicted in many locales, as are Native Americans. A fascination with people with different life-styles is evident in the images of the Shakers, the Oneida Community and the Mormons.
