Der biographische Index zu den Regesten ist ein weiterer Teil des Kommentars zu Melanchthons Briefwechsel, keine Prosopographie des 16. Jahrhunderts, aber ein Beitrag zu einer solchen. Die Auswahl der Personen ist durch ihr Erscheinen in den Regestenbänden 1-9 gegeben. Hier wird Vollständigkeit geboten. Die biographischen Angaben dienen primär dem Verständnis des Briefwechsels Melanchthons. Die Querverbindungen in diesem Corpus, insbesondere die Verwandtschaften, sind vollständig angegeben, nicht in MBW erwähnte Verwandte nur, wenn sie dem besseren Verständnis der betreffenden Person dienen, etwa ihrer sozialen Herkunft. Die dem Biogramm folgende Literatur dient dem weiteren Studium der Biographie und dokumentiert den Bekanntheitsgrad der betreffenden Person. Auf allgemeine Würdigungen wird zumeist verzichtet, insbesondere bei bekannten Personen der Reformationsgeschichte. Bei in MBW nur vereinzelt erwähnten Personen wird zur raschen Orientierung vor dem Geburtsdatum eine ganz knappe Charakterisierung geboten. Das Biogramm ist bei bekannten Personen, die in vielen Lexiken behandelt werden, kurz gehalten und beschränkt sich auf die Daten des Lebensweges. Auch Schriften werden nur ausnahmsweise zitiert, nämlich wenn sie für die Erwähnung des Autors in MBW von Bedeutung sind. Die genauen Nachweise sind im Quellenapparat der Textbände zu finden
