Localization of the Ras Isoforms May be a Contributor to Their Functional Specificity by Stephen J Lynch( Book )

We have tested the following two hypotheses: first, that N-ras and H-ras have distinct Golgi membrane localization patterns, and second, that the palmitoylation state of the Ras isoform determines both distinct Golgi membrane localization patterns as well as differences in downstream signaling of the Ras proteins. Confocal imaging revealed that H-ras and N-ras have distinct Golgi membrane localizations, and immuno-EM analyses corroborated these results. Further confocal imaging experiments determined that a singly palmitoylated H-ras mutant had a Golgi membrane localization that was similar to wildtype, singly-palmitoylated N-ras, whereas a doubly palmitoylated N-ras mutant had a Golgi localization that was similar to that of doubly palmitoylated, WT H-ras. Next, we identified factors downstream of Golgi-associated N-ras activation and tested if the palmitoylation state of the Ras isoform is important for the regulation of these genes. A microarray-based analysis identified 1652 genes that were differentially regulated by N-ras in low-level TCR-stimulated CD4+ T-cells. From these genes, 31 candidates were further tested in validation experiments, and of these, only four genes were found to have similar patterns of expression in the microarray, in empty vector and Ras isoform transductions of KO CD4+ T-cells. In the validation experiments, the expression of singly palmitoylated, WT N-ras in N-ras KO cells 'rescued' the expression pattern of the four candidate genes, whereas the expression of doubly palmitoylated Ras isoforms in KO cells did not, suggesting that the palmitoylation state of the Ras isoforms determines their ability to regulate gene expression in T-cells in the context of a low-level TCR stimulus
