Staining techniques( visu )
3 editions published in 1979 in English and held by 18 libraries worldwide
Demonstrates the preparation of bacterial cells to be stained for microscopic observation, utilizing smears made from both broth and agar slant cultures. Introduces the different types of dyes and the chemical reaction in volved in the staining process. Covers the theoretical bases and procedures for performing the Gram stain, Ziehl-Neelsen acid fast stain, and Schaeffer-Fulton endospore stain.
The care and use of the microscope( visu )
2 editions published in 1979 in English and held by 11 libraries worldwide
Introduces the parts of the compund microscope and relates their respective functions. Demonstrates the proper use of the microscope in viewing microbiological specimens, including the demonstration of total magnification for each objective lens. Suggestions for proper maintenance of the microscope are also covered.
The Making and sterilization of an inoculating loop. The development of aseptic technique. Pure culture techniques( visu )
2 editions published in 1979 in English and held by 9 libraries worldwide
The making and sterilization of an inoculating loop presents characteristics of a well-made inoculating loop compared with a variety of poorly made loops. Demonstrates method for making correct inoculating loop, and sterilization process employing a burner flame (7 min.)--The development of aseptic technique emphasizes use and development of aseptic technique as it relates to maintaining purity during transfer of stock cultures and isolated colonies to broth and agar slants (9 min.)--Pure culture techniques defines concept of a pure culture and demonstrates two isolation tecniques: streak plate and loop-dilution pour plate. Depicts preparation of an agar plate in conjunction with streak plate technique (16 min.).
Miscellaneous biochemical tests IMViC reactions( visu )
2 editions published in 1979 in English and held by 8 libraries worldwide
(Critical) This program demonstrates various biochemical tests for bacteria identification: catalase, oxidase, nitrate reduction, ammonification, urease, indole, methylred, voges-proskauer, and citrate. Reactions, underlying principles, and specific techniques are discussed. False positive or negative results are not considered; outdated procedures are included; a paucity of detail is presented; and excessive time is awarded repetitive procedures. The color quality is poor, significantly impairing the educational value of this material. Although the methodology is generally current and the explanations adequate, this is a slow, weak, vague presentation which is not recommended for use.
Water analysis( visu )
2 editions published in 1979 in English and held by 7 libraries worldwide
(Critical) This program demonstrates the performance of coliform tests on potable water. The Most Probable Number (MPN) and the Millipore Filter techniques are illustrated, and typical water purification process and collection of the water sample for analysis are detailed and rationalized. Demonstrations of presumptive, confirmed, and complete tests are presented. The instructional format is clear and effective, provides good reinforcement of concepts, and is of appropriatelength; however, the visualization could be enhanced by appropriate use of color. The program is recommended for nondegree and undergraduate allied health programs.
Fermentation reaction( visu )
2 editions published in 1979 in English and held by 7 libraries worldwide
Demonstrates the methods of performing and reading fermentation reactions using pH indicators, litmus milk (along with other observations), Kligler iron agar, and TSI agar. Includes hydrogen sulfide production using the latter media along with peptone iron agar and lead acetate agar. Reviews methods of performing and reading tests for starch hydrolysis, lipid hydrolysis, casein hydrolysis, hemolysis production and gelatin hydrolysis.
Standard plate count of pasteurized and raw milk( visu )
2 editions published in 1979 in English and held by 7 libraries worldwide
Covers the standard technique for determining viable bacterial populations in both pasteurized and raw milk samples. Covers identification of general sources of milk contamination and which diseases can be transmitted to man by drinking contaminated milk, serial dilution, plating and growth of yogurt bacteria as well as an examination of some distinctive cellular characteristics.
Testing of antimicrobial agents( visu )
2 editions published in 1979 in English and held by 5 libraries worldwide
Presents the basic techniques employing filter paper discs to determine antimicrobial effectiveness of disinfectants/antiseptics and antibiotics/sulfonamides. Identifies and measures zones of inhibition.
The Making and sterilization of an inoculating loop( visu )
2 editions published in 1979 in English and held by 5 libraries worldwide
Presents characteristics of a well-made inoculating loop and compares them with poorly-made loops. Demonstrates making a correct loop along with the burner sterilization method.
Bacterial motility( visu )
1 edition published in 1979 in English and held by 5 libraries worldwide
This program presents film footage of various types of microbial motility together with the procedure for preparation of a wet mount to observe bacteria and other microorganisms. The flagella stain and soft agar tube meth od are demonstrated to show bacterial motility.
