All-trans-retinoic acid (atRA), an active metabolite of vitamin A, regulates embryonic patterning, cellular homeostasis, and cell survival, differentiation, growth and apoptosis. atRA effects are largely mediated by retinoic acid receptors (RARs), a class of nuclear receptor generally believed to function exclusively in transcription. Analysis of vitamin A-deficient rodents reveals that adequate atRA signaling also is necessary for normal hippocampus-dependent learning and memory formation, and for synaptic plasticity underlying these processes. The mechanism(s), however, remain obscure. Using a reduced preparation of cultured hippocampal neurons, I show that atRA rapidly induces dendritic growth and translation, culminating in spine formation. In synaptoneurosomes, atRA activates mitogen-activated protein kinase (MAPK) and mammalian target of rapamycin (mTOR) signaling, both of which are required for dendritic translation activation. A membrane-associated RARalpha, revealed by surface biotinylation and live neuronal staining, mediates these rapid, non-genomic effects, siRNA knockdown of RARalpha abolished atRA-induced growth and reduced spines. I used immuno-isolated heterologously-expressed RARalpha as the bait to precipitate its associated complex(s) from hippocampal neuronal lysates to understand the molecular basis for RARalpha action. Mass spectrometric analysis of the precipitated complex unveiled the interaction of RARalpha with many RNA-binding proteins implicated in dendritic mRNA transport in neuronal RNA granules. Dendritic mRNAs are held dormant until they reach sites of action (e.g. underneath the synapses), where neuronal activity instructs their release and translational activation. A tethering assay revealed that RARalpha suppresses translation of associated mRNA. Polysome profiling indicated that RARalpha inhibits translation initiation. Recombinant RARalpha also repressed translation independent of the mRNA sequence and modifications, suggesting general translation repression. Consistent with the observation that atRA signaling specifically modifies dendritic translation and surface expression of the AMPA receptor subunit 1 (GluR1), an RARalpha antibody specifically precipitated GluR1 mRNA, among other dendritic transcripts. In situ hybridization further supported GluR1 as an mRNA cargo in RARalpha ribonucleoparticles. Sucrose gradient analysis demonstrated that a small fraction of RARalpha (possibly modified forms) cofractionated with polysomes, suggesting that RARalpha may regulate translation bidirectionally. These results reveal the surprising complexity of nuclear receptor function, demonstrating that translational, in addition to transcriptional, mechanisms mediate atRA-dependent synaptic modification
