No presente trabalho se trata da ocorrencia do agente fixador de nitrogenio no solo, o azotobacter, na area do Centro Nacional de Ensino e Pesquisas Agronomicas, situada na Baixada de Sepetiba. Foram examinadas 27 diferentes amostras pelo metodo de meio liquido e pelo metodo de Winogradsky usando placas de silica-gel. Embora 18 destas amostras tivessem um pH abaixo de pH 6 e tres abaixo de pH 5, foi possivel isolar azotobacter de 22 destes 27 solos. Os solos mais acidos nos quais foram encontradas esta bacteria tiveram o pH 4, 85 - 4,82 e 4,62. as tribos isoladas puderam ser identificadas como Azotobacter chroococcum Beijerinck com grandes celulas, aos pares, e com pigmento castanho-escuro, nao se tratando, pois de Azotobacter indicum Starkey, isolado por STARKEY em solos acidos da Malaia, o qual se caracteriza por crescimento muito lento, por celulas pequenas e por ausencia de qualquer pigmento. Em conseqüência da estufa, foi estudado, em Controsema pubescens Benth, o efeito da calagem e do revestimento calcario da semente sobre a fixacao simbiotica de nitrogenio, em solo que apresentava toxidez de manganes. Estudou-se tambem o efeito de altas temperaturas de solo na fixacao do nitrogenio. A calagem que eliminou a toxidez de manganes, aumentou de 65% a quantidade de nitrogenio fixado. O envoltorio de semente com calcario, apesar de nao ter reduzido a absorcao do manganes, aumentou o numero de nodulos, tanto quando a calagem. O calcio fornecido no envoltorio da semente nao teve, porem, efeito na fixacao de nitrogenio em si, que foi afetada, nesse caso, pela toxidez de manganes. Altas temperaturas do solo, acima de 30oC, prejudicaram a nodulacao, a fixacao de nitrogenio e a producao de forragem. A adubacao nitrogenada nao aumentou a producao de Centrosema, a nao ser nas testemunhas nao inoculadas

Apresentacao de metodos de isolamento e caracterizacao dos principais grupos de bacterias diazotroficas aerobias e endofitas, ate entao desconhecidos. Sao mencionados, tambem, os metodos de esterilizacao de raizes, folhas, colmos e sementes, normalmente utilizados nos estudos de isolamento de bacterias diazotroficas. Constam, ainda, relatos sobre o metodo de contagem em placas e meio de cultura semi-solido, assim como os metodos para manutencao dos isolados. Por fim, sao apresentadas algumas sugestoes de como proceder para o isolamento e identificacao de novas bacterias diazotroficas

O objetivo deste trabalho e estudar a acao da sacarose na microflora e, em particular, os efeitos dessa microflora na fixacao do nitrogenio e na agregacao do solo. Varios autores tem estudado as influencias da microflora nas caracteristicas fisicas e quimicas do solo, mas no Brasil poucos estudos tem sido realizados. Myers e Jones fizeram experimentos que demonstraram que a sacarose nao tem efeito direto sobre o solo.Estes autores, tambem Peele, mostraram que a sacarose atual indiretamente, como fonte de energia disponivel para o desenvolvimento da microflora. Ela e rapidamente decomposta em CO2 e H2O e nao deixa apreciaveis quantidades de residuos organicos, alem dos produtos de secrecao do material celular dos microorganismos

Herbaspirillum-like bacteria in banana plants by Olmar B Weber( )

Diazotrophic bacteria isolated from banana plants were characterized by morphological and physiological aspects. Three different groups of these plant-bacteria could be established. Two of them showed similarity to species of the Herbaspirillum genus. The third one was different because used only a few carbon substrates and produced water diffusible compounds that fluoresced under UV light. All three bacterial groups were thin rods with mono or bipolar flagella, presented negative reaction in Gram stain, showed catalase activity, were able to reduce nitrate and grew better in semi-solid JNFb medium at 31[vbar]C. The nitrogenase activity was detected in semi-solid N-free JNFb medium and expressed higher values when pH ranged from 6.5 to 7.0 (groups I and II) and 6.0 to 6.5 (group III). The diazotrophs isolated from banana plants were distinct from species of Herbaspirillum previously identified in gramineous plants. (Au)
