Un gruppo di scimmie antropoidi, che segnano il principio della civiltà umana, trova un monolite. Gli umanoidi vi si accostano diffidenti, poi lo toccano con singolare rispetto e qualcosa accade in loro: si accorgono per la prima volta che un osso abbandonato può diventare una clava, uno strumento utile. Si è accesa la scintilla dell'intelligenza. Nell'anno 2001, sulla Luna, un gruppo di esploratori trova un identico monolite, che denuncia un'anzianità di quattro milioni di anni e che genera attorno a sé un forte campo magnetico. Lo scienziato Heywood Floyd è incaricato di più approfondite ricerche: si pensa che il monolite racchiuda in sé il segreto del mondo, il rigenerarsi della vita stessa. Qualche tempo dopo viene inviata verso Giove un'astronave con a bordo due uomini in attività e altri tre in stato di ibernazione, oltre ad un esemplare del più perfezionato calcolatore elettronico, l' Hal 9000. Durante il viaggio solo Bowman, uno dei due astronauti, riesce a sopravvivere, poichè il computer ha causato la morte di tutti gli altri componenti l'equipaggio, volendosi sostituire all'uomo. Bowman, riuscito a penetrare nel cuore di Hal 9000, riesce a localizzare le sue funzioni cerebrali superiori e a bloccarle, venendo inoltre a conoscenza, indagando nella sua memoria, del vero scopo della missione: il monolite scoperto sulla Luna inviava un potente segnale radio verso Giove; qui forse si troverà la spiegazione dell'origine della vita. Infatti Bowman, nelle immediate vicinanze del pianeta, perde il controllo di se stesso, cadendo in un sonno vivificatore: è così che l'astronauta, superando Giove, affronta lo spazio infinito, attraversando mondi e galassie, vivendo una esaltante esperienza conoscitiva. Successivamente Bowman, in una trasposizione di immagini, si vede nella sua casa, prima in veste di astronauta, poi di maturo signore, e infine disteso sul letto in punto di morte, dove subentra il monolite, che lo fa rivivere, sotto forma di feto, in un informe bambino dagli occhi sgranati su un mondo tutto da scoprire
