(Producer) Over the past century the mouse has risen to become the premier mammalian model organism -- the stand-in for the human. Because of its short generation time and its striking genetic similarities to humans, scientists can use the mouse to study aspects of mammalian biology that have direct and immediate implications for human health. Professor Shirley Tilghman, a dynamic teacher and researcher, leads viewers through very recent advances in the study of the mouse, giving special emphasis to recent breakthroughs in the study of obesity and growth control. The program illustrates how the Human Genome Project has made possible the identification of genes that function very similarly in mice and humans. By describing the search for a gene in mice that controls weight gain, Professor Tilghman demonstrates the means by which scientists can now generate mice with mutations in specific genes, using as an example a gene that was mutated through painstaking experiments in her laboratory at Princeton University. The mutant mice were larger than normal and mimicked a human disorder called Beckwith-Wiedemann syndrome

(Producer) The nematode Caenorhabditis elegans, a microscopic worm, provides an excellent model organism to analyze development and behavior. This presentation focuses on programmed cell death. Vivid demonstrations of single-cell laser microsurgery let the viewer participate in an experiment designed to help reveal whether dying cells are "murdered" by neighboring cells or are "committing suicide." Mutations are induced and isolated, genes are characterized genetically, and DNA sequences are determined. A step-by-step series of graphic illustrations demonstrates the identification and analysis of one nematode cell-death gene, which is discovered to be similar to a human oncogene. The human oncogene is shown to be able to substitute for the nematode gene in a mutant worm. Professor Horvitz takes the viewer on a fascinating journey that unmasks the genetic basis of programmed cell death and indicates its relationship to human cancers and, perhaps, to neurodegenerative diseases such as Alzheimer's disease

(Producer) How do we identify the specific genes that control a complex process such as cell division? For the answer to this intriguing question, our program takes us to the Whitehead Institute in Cambridge, Massachusetts, where its director, Professor Gerald Fink, and his colleagues demonstrate how to isolate cell division mutants. They introduce the viewer to bakers' yeast, a single-celled eukaryote that divides rapidly and is easy to manipulate genetically. Because processes fundamental to eukaryotic cells from yeast to humans have been conserved in evolution, it is possible to learn a great deal about these functions in complex systems by studying them in yeast. Functions such as cell division, mating, and meiosis are all carried out by proteins encoded by genes. Surprisingly, mutants unable to divide provide the key link to the genes responsible for cell division. The students at the Whitehead lab perform meiotic analysis to show that the defect in the cell division mutant results from a defect in a single gene. This mutant provides the tool for isolation of the corresponding cell division gene by transformation
