This thesis is devoted to the geometry of Fano varieties and projective vector bundles over a smooth projective curve.In the first part we study the geometry of mildly singular Fano varieties on which there is a prime divisor of Picard number 1. By studying the contractions associated to extremal rays in the Mori cone of these varieties, we provide a structure theorem in dimension 3 for varieties with maximal Picard number. Afterwards, we address the case of toric varieties and we extend the structure theorem to toric varieties of dimension greater than 3 and with maximal Picard number. Finally, we treat the lifting of extremal contractions to universal covering spaces in codimension 1.In the second part we study Newton-Okounkov bodies on projective vector bundles over a smooth projective curve. Inspired by Wolfe's estimates used to compute the volume function on these varieties, we compute all Newton-Okounkov bodies with respect to linear flags and we study how these bodies depend on the Schubert cell decomposition with respect to linear flags which are compatible with the Harder-Narasimhan filtration of the bundle. Moreover, we characterize semi-stable vector bundles over smooth projective curves via Newton-Okounkov bodies

Dans cette thèse nous considérons des familles $pi : cc X to S$ de variétés compactes k"ahlerinnes au-dessus d'une base lisse $S$. Nous construisons un cône de K"ahler relatif $p : cc K to S$ au-dessus de la base de déformations. Ensuite nous démontrons l'existence des métriques hermitiennes naturelles sur les espaces totals $cc K$ et $cc X times_S cc K$ qui généralisent la métrique de Weil--Petersson classiuque associée aux familles polarisées de telles variétés. Nous obtenons aussi une métrique riemannienne sur le cône de K"ahler d'une variété compacte k"ahlerienne quelconque. Nous exprimons son tenseur de courbure à l'aide d'un plongement du cône de K"ahler dans l'espace de toutes métriques hermitiennes sur la variété. Nous démontrons aussi que si les variétés en question sont de fibré canonique trivial, alors notre métrique est la forme de courbure d'un fibré en droites holomorphe. Nous donnons ensuite quelques exemples et applications

On étudie les déformations des applications harmoniques $f$ tordues par rapport à une représentation. Après avoir construit une application harmonique tordue "universelle", on donne une construction de toute déformations du premier ordre de $f$ en termes de la théorie de Hodge ; on applique ce résultat à l'espace de modules des représentations réductives d'un groupe de Kähler, pour démontrer que les points critiques de la fonctionnelle de l'énergie $E$ coïncident avec les représentations de monodromie des variations complexes de structures de Hodge. Ensuite, on procède aux déformations du second ordre, où des obstructions surviennent ; on enquête sur l'existence de ces déformations et on donne une méthode pour le construire. En appliquant ce résultat à la fonctionnelle de l'énergie comme ci-dessus, on démontre (pour n'importe quel groupe de présentation finie) que la fonctionnelle de l'énergie est strictement pluri sous-harmonique sur l'espace des modules des représentations. En assumant de plus que le groupe soit de Kähler, on étudie les valeurs propres de la matrice hessienne de $E$ dans les points critiques

The aim of this thesis is to generalize a certain number of results of algebraic geometry to K"{a}hler geometry. We first generalize the Nadel vanishing theorem to arbitrary compact K"{a}hler manifolds. We prove also a particular version of the Kawamata-Viehweg vanishing theorem for manifolds admitting a fibration to a torus such that the generic fiber is projective. Using this result, we study the theory of deformations of compact Kähler manifolds under certain assumptions on their canonical bundles. Finally, we study varieties with nef anticanonical bundles. We prove that the slopes of the Harder-Narasimhan filtration of the tangent bundles with respect to a polarization of the form $omega_X^{n-1}$ are semi-positive. As an application, we give a simple proof of the surjectivity of the Albanese map, and we investigate also the local triviality of the Albanese map
