Abstract:

The crystal structure of porcine cathepsin H in complex with recombinant humanstefin A reveals the exopeptidase - inhibitor interaction. The Nterminal of stefin A cannot bind into the active site of cathepsin H with the same geometry as the N-terminal of stefin B in the complex with papain due to steric hindrance. It makes a beta turn and points with the Nterminal residue away from the active site cleft. C-terminal part of minichain of cathepsin H and a short insertion loop Lys155A-Asp155D are thereby displaced, their geometry appears to be disordered.
